Drug Screening with Primary Cells
Cultured primary cells possess characteristics that often represent in vivo physiology better than immortalized cell lines. Thus, primary cell-based assays could give a more accurate prediction of the activity of compounds in in vivo models and in the clinic, and thus could contribute to a decrease in attrition in the whole process of drug discovery.
Bone marrow progenitor assays are more informative than cell proliferation methods; less expensive and faster than in vivo models; and are highly reproducible. One of these assays, the CFC (colony forming cell) assay, is a more effective and cost-saving method for developing dosing strategies for Phase I clinical trials and is the only assay recognized by a regulatory body (ECVAM http://ecvam.jrc.it/) as a method to predict toxicity in phase I drug development.
